Delivering innovative technologies for peptide professionals

The global need for custom and synthetic therapeutic peptides is growing, as academic researchers and the pharmaceutical and biotech industries renew their interest in peptide-based drug discovery. As researchers require higher quality peptides in ever greater quantities and complexity, the need for reliable and innovative tools that offer greater productivity and increased performance is also growing.

Biotage is dedicated to meeting these needs and committed to developing the tools needed by the global peptide community for peptide synthesis and purification. Biotage is the only manufacturer of traditional, room temperature peptide synthesizers as well as microwave peptide synthesizers in both entry level and high throughput configurations. Biotage offers a range of manual, semi-automated and automated systems. Whether a system that makes a single peptide or multiple branched peptides is required, Biotage has an instrument to suit requirements.

Advantages of microwave assisted vs. traditional, room temperature peptide synthesis

  • Powerful technique for accelerating the synthesis of peptides
  • Rapid and precise elevation of temperature with efficient temperature control during the synthesis
  • Reduction in synthesis time and an increase in purity
  • Allows access to the synthesis of difficult peptide sequences
  • Improve coupling rates and prevent side reactions
  • Reduction in cost of synthesis by using standard coupling reagents
  • “Green Chemistry” potential, use of less harmful solvents

b-amyloid peptide is the major component of the neurological plaques in Alzheimer’s patients. Synthesis is difficult due to reported on-resin aggregation and folding due to hydrophobic C-terminus. With microwave assisted synthesis in 5 min at 75 ºC a 73% HPLC purity while with synthesis in 45 min at room temperature purity was 54%.

Peptide synthesis using microwave irradiation gives high yield and high purity compared with conventional room temperature methods. Microwave assisted peptide synthesis is also an enabling technology to produce long and/or difficult peptide sequences.
